On this road (the only fast one) you have to pay toll once, on each way, when entering Alligator Alley.

Another way, a little longer but with a different view and it may be an option if you are only going towards Naples, in Miami, go north on 8th Street ‘Calle 8’ (also known as US41). Using this road, you can get to Michigan if you want to. Obviously, with traffic, stop signs or traffic lights that may take you longer. On the way, you cross part of the Everglades until you reach Naples, continuing the road to Sarasota.
